Summary Omalizumab, a humanized anti-IgE antibody, inhibits degranulation of mast cells by direct binding to IgE.

In the beginning of this DVD, you can see development of allergic inflammation that starts from degranulation of mast cells on rat mesentery. If anti-rat IgE antibody corresponding to Omalizumab is pre-administrated, IgE-mediated inflammation dose not initiate by addition of antigen. You can notice and well understand this event by fluorescence imaging: the anti-IgE antibody seen in red effectively inhibits the binding of IgE to mast cells.

At the lung tissue of a model mouse of asthma, you observe migration of inflammation cells like eosinophils, hypertrophy of vascular intima, and increase of mucous cells. However, these inflammation phenomena are inhibited by the administration of the anti-IgE antibody.

Degranulation of human mast cells is initiated by the binding of IgE to the cells. Thus, Omalizumab effectively blocks mast cell degranulation. Moreover, you can also clearly observe that Omalizumab inhibits activation of human mast cells. This event is shown by the immunostaining of CD63, a marker protein of mast cell degranulation, which is recoded using a confocal laser scanning microscopy.
