Summary
Osteoporosis is a common disease with reducing bone mass and weakening of the bone. With the aging society in Japan, Osteoporosis has been a social problem. This film demonstrates the bone metabolism and the mechanism of osteoporosis by considering the dynamism of relating cells, osteoblasts and osteoclasts.
Bone is continually remodeled by osteoblasts which deposit fresh layers of bone and osteoclasts which erode bone matrix. In vitro culture of the bone marrow, multinucleated osteoclasts appear first, and then osteoblasts differentiate and make bone. The mesh of collagen fiber, intracellular granules and deposition of calcium phosphate are also observed.
We succeeded to film the process of making bone clearly by a long-term observation (several weeks) under a microscope and to detect for the first time in the world the key factor for bone remodeling, TGF-ΐ, in the living tissue by specific antibodies and a scanning electron microscope.
